In bio 203, we use the compound microscope. Either the Zeiss, which use here as for the purpose of demonstration, or the Nikon. With the Zeiss compound microscope, the main parts of the microscope include the ocular (or the eye pieces), where you use them to look under the microscope. You put your eyes in there. The stage where you put your specimens - so open the clip, put your microscope slide on, and gently let it clip in to secure the microscope slide onto the stage. These adjustment knobs, that have two knobs right here. One ring would move the microscope slide left and right and one up and down. So position your slide to be under the proper field of view of interest. For this microscope, we have different objectives: the 4x 10x 40x, and 100x. In bio203 we do not use the 100x objective. In order to switch from one objective to another we use revolving nose piece right here. Always start with the lowest objective and move up accordingly. For the microscope, first you want to plug your plug into power source. To turn on the microscope for the Zeiss, it's on the right-hand side. And that knob also adjusts the light intensity that goes through your slides. For the Nikon, it actually has an on and off switch just right beside the light intensity knob. There are two rings right here: The inner bigger rings are the coarse adjustment, and that will move the stage up and down. The outer smaller rings also move the stage up and down, but to the lesser extent. You only use the coarse adjustment with the 4x objective. When you have your slide secure under the microscope, first of all you use the 4x objective. Use coarse adjustment to bring everything into focus. When the image is into focus already, you can crank up to another objective to have better higher magnification. When you change objective, use fine focus knob which is again the smaller ring outside to bring your image into focus again. If the image is already into focus, you want to project them onto the screen and take the picture of the micrograph. In order to do that, remove one of the eyepiece. Keep in mind that when you remove it place that vertically into the tray of the specimens provided onto the bench so that it doesn't roll off the bench. With the eyepiece camera into the dyno light, insert your eyepiece camera onto the ocular. It should fit snugly into the ocular space. With the Dynolite usb, plug that into the usb port in the adjacent computer. In order to open the Dynoeye imaging software, first of all you log in using your utorID and password to operate the computer. From the Start menu, go to course applications dino capture 2.0. When you have your specimen in the ideal field view you would like to put scale bar into your image. To set up the scale bar, first of all, you go into the units, pick 'micron'. In the box beside that, enter the corresponding magnification according to your objective - For this one, use 4x objective, so enter 4. With the calibration, icon click on the 4x which is the corresponding magnification calibration. From that moment, you should see scale bar showing at the bottom left corner showing the proper scale bar for your image. You want to click on the snapshot icon to capture the image. Keep in mind that at the end of the day, you want to save all the images and save that on your thumb drive or email to yourself to further analyze the image down the road.
